Plant leaf parameters such as stomatal index, stomata type, microscopic features, and powder microscopy are important taxonomic used to properly identify species differentiate one from another these is a vital keys in the pharmacognostic study of medicinal plants because play role distinguishing crude drugs even when they fragmented or powdered. Also, extractive retention factor, ash value, phytochemical standardization for establishment pharmacopeia monograms. Therefore, seeks validate use phytochemistry molecular systematics among Asteraceae, Euphorbiaceae, Fabaceae support their circumscription therefore evaluated profile Desmodium velutinum, Synedrella nodiflora Mallotus oppositifolius folklorically South East Nigeria using standard laboratory protocols. The result showed that nodiflora, all had irregular shaped epidermis, was present only abaxial surface respectively. both D. velutinum S. presence lignin, starch, cystoliths, tannin, Oil body while calcium oxalate absent whereas, oxalate, cystoliths were absent. Also TLC M. 3 visible spots with factors (RF) 0.363, 0.425, 0.5 order respectively, 4 spot factor 0.5, 0.550 0.66 respectively 6 0.255, 0.438, 0.538, 0.600 0.688 also methanol exhaustible extracted phytochemicals dried plant. acute toxicity studies individually LD50 greater than 5000mg/kg. This validates claim identification drugs.
